The Bioaim Human DAN ELISA kit is a solid phase sandwich ELISA (enzyme-linked immunosorbent assay) for the quantitative measurement of DAN in Human serum, plasma and cell culture media. An antibody specific for Human DAN was coated on a 96-well plate. Standards and samples are added to the wells and any DAN present binds to the immobilized antibody. The wells are washed and biotinylated anti-Human DAN antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is added to the wells. The wells are again washed and TMB substrate solution is added, which produces a blue color in direct proportion to the amount of DAN present in the initial sample. The Stop Solution changes the color from blue to yellow, and the microwell absorbances are read at 450 nm.

Background: DAN has now been shown to be a prototypical member of the DAN family of secreted glycoproteins that are putative antagonists for TGF-beta superfamily proteins. DAN family members share a cysteine-rich domain that is structurally related to the cysteine-knot motif found in TGF-beta superfamily ligands. There are at least five mammalian DAN family members including DAN, Gremlin/DRM, Cer1 , Dante and PRDC .
