The Bioaim Human CEACAM-1 ELISA kit is a solid phase sandwich ELISA (enzyme-linked immunosorbent assay) for the quantitative measurement of CEACAM-1 in Human serum, plasma and cell culture media. An antibody specific for Human CEACAM-1 was coated on a 96-well plate. Standards and samples are added to the wells and any CEACAM-1 present binds to the immobilized antibody. The wells are washed and biotinylated anti-Human CEACAM-1 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is added to the wells. The wells are again washed and TMB substrate solution is added, which produces a blue color in direct proportion to the amount of CEACAM-1 present in the initial sample. The Stop Solution changes the color from blue to yellow, and the microwell absorbances are read at 450 nm.

Background: Carcinoembryonic Antigen-related Cell Adhesion Molecule 1 (CEACAM-1), also known as CD66a and BGP-I, is a 115 kDa type I transmembrane glycoprotein that belongs to the membrane-bound CEA subfamily of the CEA superfamily. On the surface of cells, CEACAM-1 forms non-covalent homo- and heterodimers. The extracellular region contains three C2-type Ig-like domains and one N-terminal V-type Ig-like domain. The encoded protein mediates cell adhesion via homophilic as well as heterophilic binding to other proteins of the subgroup. Multiple cellular activities have been attributed to the encoded protein, including roles in the differentiation and arrangement of tissue three-dimensional structure, angiogenesis, apoptosis, tumor suppression, metastasis, and the modulation of innate and adaptive immune responses. Multiple transcript variants encoding different isoforms have been reported, but the full-length nature of only two has been determined.
