The Bioaim Human CD80/B7-1 ELISA kit is a solid phase sandwich ELISA (enzyme-linked immunosorbent assay) for the quantitative measurement of CD80/B7-1 in Human serum, plasma and cell culture media. An antibody specific for Human CD80/B7-1 was coated on a 96-well plate. Standards and samples are added to the wells and any CD80/B7-1 present binds to the immobilized antibody. The wells are washed and biotinylated anti-Human CD80/B7-1 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is added to the wells. The wells are again washed and TMB substrate solution is added, which produces a blue color in direct proportion to the amount of CD80/B7-1 present in the initial sample. The Stop Solution changes the color from blue to yellow, and the microwell absorbances are read at 450 nm.

Background: Cluster of differentiation 80 (also CD80 and B7-1) is a protein found on activated B cells and monocytes that provides a costimulatory signal necessary for T cell activation and survival. It is the ligand for two different proteins on the T cell surface: CD28 (for autoregulation and intercellular association) and CTLA-4. CD80 works in tandem with CD86 to prime T cells. Although both CTLA-4 and CD28 can bind to the same ligands, CTLA-4 binds to B7-1 and B7-2 with a 20 – 100 fold higher affinity than CD28 and is involved in the down-regulation of the immune response. B7-1 is expressed on activated B cells, activated T cells, and macrophages. B7-2 is constitutively expressed on interdigitating dendritic cells, Langerhans cells, peripheral blood dendritic cells, memory B cells, and germinal center B cells
