The Bioaim Human CD163 ELISA kit is a solid phase sandwich ELISA (enzyme-linked immunosorbent assay) for the quantitative measurement of CD163 in human serum and plasma. An antibody specific for human CD163 was coated on a 96-well plate. Standards and samples are added to the wells and any CD163 present binds to the immobilized antibody. The wells are washed and biotinylated anti-human CD163 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is added to the wells. The wells are again washed and TMB substrate solution is added, which produces a blue color in direct proportion to the amount of CD163 present in the initial sample. The Stop Solution changes the color from blue to yellow, and the microwell absorbances are read at 450 nm.

Background: CD163 is the high affinity scavenger receptor for the hemoglobin haptoglobin complex and in the absence of haptoglobin – with lower affinity – for hemoglobin alone. It has also been shown to mark cells of monocyte/macrophage lineage. CD163 mediates monocyte binding to bacteria, leading to the release of inflammatory cytokines. It can also mediate monocyte-erythroblast adhesion and promote erythroblast expansion. CD163 binds and internalizes the cytokine TWEAK, and the ratio of soluble CD163 to TWEAK in the plasma is elevated during atherosclerosis.
