The Bioaim Human ANGPTL4 ELISA kit is a solid phase sandwich ELISA (enzyme-linked immunosorbent assay) for the quantitative measurement of ANGPTL4 in Human serum, plasma and cell culture media. An antibody specific for Human ANGPTL4 was coated on a 96-well plate. Standards and samples are added to the wells and any ANGPTL4 present binds to the immobilized antibody. The wells are washed and biotinylated anti-Human ANGPTL4 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is added to the wells. The wells are again washed and TMB substrate solution is added, which produces a blue color in direct proportion to the amount of ANGPTL4 present in the initial sample. The Stop Solution changes the color from blue to yellow, and the microwell absorbances are read at 450 nm.Research Area Angiogenesis;
angiopoietin like 4(ANGPTL4) Background: Angiopoietin-like, also known as angiopoietin-related proteins, are structurally related to the angiopoietins. Seven ANGPTL proteins have been identified (ANGPTL1 through 7). ANGPTL4 plays an important role in numerous cancers and is implicated in the metastatic process by modulating vascular permeability, cancer cell motility and invasiveness. ANGPTL4 contributes to tumor growth and protects cells from anoikis, a form of programmed cell death induced when contact-dependent cells detach from the surrounding tissue matrix. ANGPTL4 secreted from tumors can bind to integrins, activating downstream signaling and leading to the production of superoxide to promote tumorigenesis. ANGPTL4 disrupts endothelial cell junctions by directly interacting with integrin, VE-cadherin and claudin-5 in a sequential manner to facilitate metastasis. ANGPTL4 functions as a matricellular protein to facilitate skin wound healing.
