The Bioaim Human 4-1BB ELISA kit is a solid phase sandwich ELISA (enzyme-linked immunosorbent assay) for the quantitative measurement of 4-1BB in Human serum, plasma and cell culture media. An antibody specific for Human 4-1BB was coated on a 96-well plate. Standards and samples are added to the wells and any 4-1BB present binds to the immobilized antibody. The wells are washed and biotinylated anti-Human 4-1BB antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is added to the wells. The wells are again washed and TMB substrate solution is added, which produces a blue color in direct proportion to the amount of 4-1BB present in the initial sample. The Stop Solution changes the color from blue to yellow, and the microwell absorbances are read at 450 nm.Research Area Apoptosis;
Tumor necrosis factor receptor superfamily 9(4-1BB) Background: 4-1BB, also known as CD137 and ILA, is a TNF receptor superfamily member and has been designated TNFRSF9. 4-1BB can be expressed by activated T cells, but to a larger extent on CD8 than on CD4 T cells. In addition, CD137 expression is found on dendritic cells, follicular dendritic cells, natural killer cells, granulocytes and cells of blood vessel walls at sites of inflammation. It is also expressed on monocytes, neutrophils, DCs and eosinophils. The ligand for 4‑1BB (4‑1BBL), also named TNFSF9, belongs to the TNF ligand superfamily. 4‑1BBL is predominantly expressed on activated antigen presenting cells such as B cells, macrophages and dendritic cells. It is also expressed on most T and B lymphoma cell lines. In response to 4‑1BBL binding, 4‑1BB transduce a T cell costimulatory signal in both CD4+ and CD8+ T cells to promote survival and enhance proliferation, cytokine production and effector function.
