The Bioaim Canine Resistin ELISA kit is a solid phase sandwich ELISA (enzyme-linked immunosorbent assay) for the quantitative measurement of Resistin in canine serum, plasma and cell culture media. An antibody specific for canine Resistin was coated on a 96-well plate. Standards and samples are added to the wells and any Resistin present binds to the immobilized antibody. The wells are washed and biotinylated anti-canine Resistin antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is added to the wells. The wells are again washed and TMB substrate solution is added, which produces a blue color in direct proportion to the amount of Resistin present in the initial sample. The Stop Solution changes the color from blue to yellow, and the microwell absorbances are read at 450 nm.Research Area Peptide Hormone; Lipid metabolism;
Resistin (Resistin ) Background: Resistin, also known ADSF and FIZZ3, is a secreted peptide hormone that circulates as either a noncovalent trimer, or a disulfide-linked homohexamer. Resistin is expressed by white adipocytes, splenocytes, astrocytes, and anterior pituitary epithelium. It blocks Insulin-stimulated uptake of glucose by adipocytes and promotes glucose release by hepatocytes. High circulating Insulin levels induce Resistin secretion which then antagonize Insulin action. resistin has been shown to increase transcriptional events, leading to an increased expression of several pro-inflammatory cytokines including (but not limited to) interleukin-1 (IL-1), interleukin-6 (IL-6), interleukin-12 (IL-12), and tumor necrosis factor-α (TNF-α) in an NF-κB-mediated (nuclear factor kappa-light-chain-enhancer of activated B cells-mediated) fashion. It has also been demonstrated that resistin upregulates intercellular adhesion molecule-1 (ICAM1) vascular cell-adhesion molecule-1 (VCAM1) and chemokine (C-C motif) ligand 2 (CCL2), all of which are occupied in chemotactic pathways involved in leukocyte recruitment to sites of infection. Resistin itself can be upregulated by interleukins and also by microbial antigens such as lipopolysaccharide, which are recognized by leukocytes.
